活性化NKT細胞によるがん転移抑制機構と臨床応用に関する基礎研究

研究概要	本年度の研究実績は次の2点である。 1)NKT細胞上の腫瘍細胞を認識するレセプター分子に関する解析:NKT細胞が腫瘍細胞を傷害する際、腫瘍細胞を認識する活性化および抑制性のレセプターは不明である。これまでに樹立したモノクローナル抗体やサブトラクション法(NKT細胞-T細胞)を用いてこれらのレセプターをクローニングし、新規分子NKG2Iを同定した。その機能をin vitro及びin vivoで解析した。NKG21はCD94のホモログで、抗体で架橋刺激するとサイトカイン産生が上昇し、in vivoの解析からはアロの細胞の拒絶反応に関与していることがわかった。 2)NKT細胞活性化を目的とした免疫細胞療法に関する研究。これまでに、患者単核球をGM-CSFとIL-2を用いて、GMP基準とFDA細胞治療ガイドラインに沿った形で培養し、in vitroで十分なαGalCerの抗原提示能を誘導する培養系を樹立した。これを用いてphase Iの研究を行った。実際に治療を受けた患者での内在性のNKT細胞(非常に少なく機能評価が困難)の活性化を評価するために、single cell sortingとsingle cell PCRの系を樹立した。現在までに11例のPhaseI相当の研究を行ったが、重篤な有害事象はでていない。ヒトへの臨床応用に関しては、千葉大学の倫理委員会で承認されたプロトコールを遵守して、PhaseIの臨床研究を行った。現在、新たに倫理委員会で承認されたプロトコールに即して、PhaseIIの研究のための準備を行っている。最終的には、肺がんの手術後の微小転移をターゲットにして、がんペプチドを用いたCTL療法との併用を目指す。
