消化管癌の分子病理診断用ミニcDNAアレイ及び抗体アレイの開発と実践導入

研究概要	消化管癌のマイクロアレイ技術を用いた分子病理診断による癌の個性の実践導入をを目指して、次のことを行った。 1)消化管癌について、serial analysis of gene expression(SAGE)法を用いて網羅的遺伝子発現解析を行った。胃の高分化腺癌、低分化腺癌及び非癌部胃粘膜を対象に解析し、generating gene type IV(Reg IV)をはじめ胃癌に特異的に高発現する種々の遺伝子が同定された。Reg IVは、realtime RT-PCRによる検索では確かに胃癌で高発現しており、ステージと相関していた。さらに、それぞれの組織型に特異的発現を示す遺伝子がいくつか同定され、それには、細胞増殖、染色体安定性、蛋白分解などに関わるものが含まれていた。胃癌特異的に発現していた数種のtagは全く未知のものであり、reverse SAGEにより遺伝子の同定を行っている。 2)胃癌患者及び健常人ボランティアを対象とし、遺伝子多型(SNP)と癌の発生・進展との関連を解析した。E-cadherinでは、rSNPとリンパ節転移との間に相関傾向がみられ、MMP-1では、rSNPと胃癌の発生及びステージとの間に有意な相関が認められた。 3)診断用ミニcDNAアレイを用いた分子病理診断システムを構築した。ホルマリン固定あるいは新鮮凍結された生検組織について、laser microdissectionを併用して正確にDNA及びRNAを抽出し、それぞれの臓器癌に対する診断用ミニcDNAアレイを用いて癌の個性診断を行うとともに、正常粘膜試料では、消化管癌に関連したSNP同定用ミニアレイを使用し、発癌感受性、薬剤感受性、遺伝性腫瘍の同定などを行うものである。この実践運用により、新しい癌の予防・診断・治療の展開が期待できる。 4)蛋白アレイ作成のための蛋白の固相化及び抗原・抗体反応の成立の条件設定を行っており、実践導入は今後の課題である。
