樹状細胞の遺伝子操作によるがん免疫強化法に関する研究

研究概要	ヒト末梢血単球よりGM-CSFとIL-4を用いて誘導した樹状細胞を用い、ヒト樹状細胞への遺伝子導入法の確立を目指した研究を行った。まずアデノウイルスベクターAxCAEGFPを作成し遺伝子導入効率を検討した。通常の感染方法では、アデノウイルスを用いた場合においても高い効率で遺伝子導入するには高濃度のウイルスが必要であった。一方、アデノウイルス感染時に37℃で2000xg、2時間の遠心を併用することで10〜50MOIでも遺伝子導入効率は80%以上に向上すると共に、遺伝子発現強度も有意に増加した。また、本法で遺伝子導入した樹状細胞の生存率は遺伝子導入をしていない樹状細胞のそれと同等で、混合リンパ球反応におけるリンパ球増殖刺激効果も保たれており、樹状細胞へのダメージは認められなかった。そこでアデノウイルスベクターを用いてヒト単球由来樹状細胞へのインターロイキン12(IL-12)遺伝子導入を行い、その免疫学的性状を解析した。我々の開発した遠心法を用いてAxlCihp40ip35を感染させたところ、経時的にIL-12p70の産生が認められ、48時間で8.lng/10*6cellsのIL-12産生を得た。IL-12遺伝子導入樹状細胞の表面抗原発現では、コントロールのEGFP遺伝子を導入した樹状細胞と比較してCD80、CD83、CD86、HLA classI、classIIの発現増強を認め、やや成熟した表現形を示した。しかし、FITC-デキストランを用いた貧食能の検討ではコントロールの樹状細胞と同程度の貧食能を認めた。またIL-12遺伝子導入ヒト樹状細胞の免疫増強効果を、アロ及びオートのヒト混合白血球反応を用いて検討した。その結果、いずれの反応においてもIL-12遺伝子導入樹状細胞は、コントロールのEGFP遺伝子導入樹状細胞に比較して、リンパ球刺激活性の著しい増強を認めた。以上からヒト樹状細胞に対してもIL-12遺伝子導入が可能であり、IL-12遺伝子導入ヒト樹状細胞は強力な免疫増強作用を有することが示唆された。
