研究概要	近年、癌化学療法においては新しい分子標的の探索が薬剤開発のブレークスルーとなることから、様々な視点から薬剤感受性規定分子の同定が進められている。固形癌治療で最も有力な抗癌剤であるシスプラチンは、DNA障害のシグナル伝達を細胞に惹起する。この経路に登場する感受性を規定する新しい分子を見いだし、固形癌の分子標的としての評価を試みる。我々は以前より損傷DNA認識蛋白としての機能を持つ転写因子及びDNA障害で活性化される転写因子に着目していた。本研究において特にシスプラチン修飾DNA認識蛋白群のDNA損傷シグナルにおける役割を明らかにする目的で本年度は以下の点を明らかにした。 1 YB-1は一本鎖核酸に強く結合し、3'→5'exonuclease活性を持つなど特異な性質を見いだした。 2 YB-1の臨床試料での発現を大腸癌や肺癌で検討し、悪性度や予後との関連を明らかにした。 3 HMG1蛋白の損傷DNA認識活性をp53が増強することを見いだした。 4 耐性細胞の解析から、DNA複製に関与する転写因子CTF2がHMG1の発現にも関与することを見いだした。 5 シスプラチン誘導遺伝子としてディファレンシャルディスプレイ法により転写因子ATF4を同定した。その発現は耐性細胞で高発現していることから、シスプラチン感受性との関連が推測された。現在、p53/p73癌抑制遺伝子産物と転写因子との分子会合の生物学的意義についても解析を進めており、分子会合阻害による分子標的治療を考案したい。
