ビタミンE転送蛋白質遺伝子と発がんリスクの個体差に関する分子疫学的研究

研究概要	ビタミンE転送蛋白質(α-Tocopherol Transfer Protein;α-TTP)遺伝子の多型と、血中ビタミンE濃度およびがんを含めた疾患発症との間の相関を検討した。まず、互いに強い連鎖不平衡にある頻度の高い2つの遺伝子多型を同定した。イントロン4の5'端より11番目の位置におけるCとTの一塩基多型に関しては、日本人対照群(n=300)におけるC-アリル/T-アリルの頻度がそれぞれ0.76/0.24であるのに対し、冠動脈疾患患者群(n=150)では0.73/0.27、肺癌患者群(n=150)では0.75/0.25と差を認めなかったが、膵臓癌患者群(n=130)ではそれぞれ0.68/0.32と、T-アリルの頻度が有意に高い傾向が認められた(p<0.02)。次に、150名のボランティアを対象として調べたところ、有意差には至らないが、遺伝子型がC/C、C/T、T/Tの順に血中ビタミンE濃度が低下する傾向が見られた。一方、α-TTP遺伝子の転写開始地点より数えて335塩基5'上流地点に、アデニン(A)が9-12回繰り返す[-335A(n)多型]を同定した。24名の日本人における、-335A(n)多型のA(10)-アリルとA(11)-アリルの各対立遺伝子頻度はそれぞれ、0.71と0.29であった。イントロン4多型と-335A(n)多型の遺伝子型は、24人中22人で完全に一致しており、強い連鎖不平衡が認められた[T-アリルはA(11)-アリルと連鎖]。293T細胞を用いた感染発現実験により検討した結果、A(11)に比べてA(10)の方が有意に(p<0.05)転写活性化に与える影響が強いことが明らかとなった。最後に、糖尿病患者を対象とした解析では、糖尿病患者群において遺伝子型がC/Cの者では、γ-tocopherol濃度が有意な低下を示した(p=0.01)。
