単純ヘルペスウイルスの病原性発現におけるアクセサリー遺伝子の役割

研究概要

単純ヘルペスウイルス(HSV)は少なくとも74種の遺伝子をコードするが、その半数以上は培養細胞での増殖には必須ではないアクセサリー遺伝子であることが明らかにされている。本研究はHSVの病原性発現におけるアクセサリー遺伝子の役割、作用機構を明らかにしようとするものである。マウスに対する病原性発現におけるUL56の重要性についてはBeckerのグループによる一連の研究があるが、その役割や機能については全く分かっていないといって良い。我々もUL56欠損ウイルスの病原性については、その特性を確認したが、特異抗体、真核発現系などを用いた解析から、この蛋白質が小胞輸送に関わることを示唆する結果を得た。
UL56遺伝子は、塩基配列より235個のアミノ酸からなる蛋白質をコードしていることが予想された。大腸菌における発現系を作製し、精製したGST-UL56融合蛋白質を用いて特異抗体を作製した。この抗体はHSV2型感染細胞の35,34,33,32,31及び30kDa蛋白質と特異的に反応した。Vero細胞でのUL56単独発現では31、30kDaの蛋白質のみを検出した。HSV2型では1型と異なり、開始コドンの10アミノ酸上流部位にATGが存在する。上流部を含む発現系を作製し検討したところ、34,32,31,30 kDa、の4本のバンドが検出された。以上の結果は、上流のATGからも翻訳が開始されていることを示唆するものである。また、蛍光抗体法による観察では感染6時間後より検出され、ゴルジ体のマーカーGolgi58KProteinと共局在した。より感染の後期ではUL56蛋白質は核周囲の部に集積すると同時に、細胞質にも顆粒状に検出された。単独発現では、ゴルジ体への集積像とともに細胞質に顆粒状に分布し、後者はearly endosomeのマーカーEEA1と共局在した。さらに、界面活性剤に対する挙動、sucrose gradient fractionationによる解析の結果から、UL56蛋白質はlipid raftsと会合していることが示唆された。
