ウイルス誘導細胞融合制御因子FRP-1複合体の構造と機能

研究概要	FRP-1 knockout miceの作製:CD98ノックアウトマウスの作製は常法に従った。一方のCD98遺伝子を欠損したヘテロマウス(+/-)においては外見上異常は認められなかった。ヘテロマウスどうしの掛け合わせによって、CD98遺伝子を欠損したホモマウス(-/-)を得ようとしたが、生まれてくるマウスの中にはホモマウスは認められなかった。胎生期のどの段階で致死に陥っているか検討したところ、妊娠初期の段階であるブラストシスト(3.5日)までにCD98遺伝子を欠損したホモマウスは死亡する事が明らかになった。 FRP-1媒介性細胞融合制御におけるADAM-9の関与:血液単球に抗FRP-1/CD98抗体を作用させると、多核巨細胞が形成され、さらに培養すると破骨細胞に分化する。何故、血液単球はウイルスの関与無しに、抗FRP-1/CD98抗体刺激によって、細胞融合が生じるのだろうか?血液単球にはウイルス糖タンパクに匹敵するものが発現しているのであろうか?この問題意識のもとに血液単球の多核巨細胞が形成におけるADAM Familyの役割を解析した。 (i)血液単球に抗FRP-1/CD98抗体やRANKL+M-CSPを作用させて、ADAM Family mRNAの発現の変化を解析したところ、ADAM9&10mRNAの発現が特異的に増強した。また血液単球を抗FRP-1/CD98抗体で刺激すると、ADAM9&10が発現してくる事を見出した。 (ii)FRP-1/CD98媒介性細胞融合とRANKL媒介性細胞融合は抗ADAM9抗体や抗ADAM10抗体によって阻害される。 (ii)FRP-1/CD98媒介性細胞融合とRANKL媒介性細胞融合はADAM9阻害剤によって抑制される。 FRP-1のlight chainのN末端半分と高いホモロジーを有する蛋白(N-light chain):FRP-1のlight chainのN末端側と高いホモロジーを有する蛋白を3つ同定した。
