HIV外皮蛋白断片gp41-C34を基にした人工設計型細胞融合阻害剤の創製研究

研究概要	HIVの外被蛋白質gp41には膜融合成立に必須な2ヶ所のα-helix部位を含む。すなわち、gp41の細胞外ドメイン中、そのN端側(N-region)およびC端側(C-region)に存在する2つの領域である。膜融合時には、N-regionはtriple helixを形成し、引き続きC-regionがhelixを形成してN-regionのtriple helixを取り囲み、アンチパラレルな6-helical bundle構造が形成され、その結果、ウイルス側と標的細胞側の膜が近付き、膜融合が成立するものと推定されている。従って、この6-helical bundle構造形成阻害剤創製のため、我々は、C-regionの34残基のペプチド(C34)を基盤とし、C34ペプチドの臨床応用を妨げていると考えられる難溶性を克服し、かつその抗HIV活性を向上させたペプチド(Soluble C34;SC34)のde novoデザイン、合成を行った。C34は、N-region由来のN36ペプチドと相互作用し、前述の6-helical bundle構造を取ることがX線結晶解析より明らかにされている。そこで、C34とN36の相互作用様式を参考に以下のコンセプトによるSC34のデザインを行った。 (1)C34のN36との相互作用部位は既存の配列とする。 (2)6-helical bundle構造の外側に位置する溶媒接触領域(b, f, c, g位)には側鎖間salt bridge形成によるα-helicity及び水溶性の向上を目的に、i-i+4位にGlu及びLys置換を導入する。以上のコンセプトに基づき合成したSC34は、C34に較べ大幅な水溶性の向上が見られ、さらにその抗HIV活性を測定したところ現在phaseIII臨床試験中であるC-peptideの1つT-20の約10倍、C34とほぼ同等の活性を示すことが明かとなった(IC_<50>=1.0nM)。また、SC34は、T-20耐性HIV株にも有効であった。これらの結果より、我々のデザインコンセプトは、HIV-cell fusion inhibitorの開発に有用なものであると判断した。
