粘膜ワクチンへの基礎研究:HIV特異的分泌型IgA誘導型アジュバント開発

研究概要

粘膜免疫システムを駆使してHIVに対する第一線のバリアを構築する為に、経口・経鼻ワクチンが有効であり、その鍵を握っているのが粘膜免疫増強効果のある粘膜アジュバントの開発である。そこで、本研究計画ではHIV粘膜ワクチンに向けて新規アジュバント開発の為の基礎研究を展開している。新規粘膜アジュバントとして、我々が開発したmCT、E112Kの免疫増強作用の有効性をさらに改良する為に、B. choshinensis発現ベクターを使ってE112Kのm112KのmCT-Aサブユニットと病原性大腸菌由来易熱性毒素のLT-Bサブユニット遺伝子(pNCMO2-LTB-mCTA)を発現しているキメラ型アジュバントの作成に成功している。つまり、CT-Bに比較してLT-Bが宿主細胞の発現する多数のリガンド分子(GM1,asialoGM1,GM2)に結合できる特徴をいかして、粘膜上皮細胞へのワクチン抗原などの送達効果増強が期待できる。Bc-mCT-A/LT-Bキメラ型アジュバントの安全性と免疫増強効果はin vivo, in vitro実験系両方を駆使して検討した。毒性試験に関しては、腸管傑紮ループ試験、cAMP誘導性試験、ADPリボシルトランスフェラーゼ試験、CHO試験を併用してその安全性を確認することができた。さらに、タンパク性ワクチン抗原とキメラ型を混合して経鼻免疫するとワクチン特異的IgAとIgG抗体が粘膜系と全身系免疫システム両方に誘導された。次に、ワクチン抗原としては霊長類でのSHIVを使った感染防御免疫誘導実験を踏まえて、HIVgp160,env, p55などのワクチン抗原としてBc-mCT-A/LT-B(またはmCT)と混合したものを実験用マウスに経鼻・経口投与してHIV特異的免疫応答の誘導を粘膜系と全身系免疫機構で比較・検討する実験を現在進めている。特にHIV抗原特異的分泌型IgA (例:生殖器や腸管分泌液、唾液)と血清IgGサブクラス抗体の誘導とその中和効果を検討している。さらに新規ハイブリッド型FLのHIV経鼻ワクチンへの応用についても検討中である。
