デングウイルスに対する宿主受容体の同定と感染制御

研究概要

1.デングウイルス感受性細胞の簡便な測定法を開発した。
標的細胞に対してウイルス液を反応させ、細胞に吸着したウイルスをウイルス抗体、およびフローサイトメータにより検出する新しい簡便な方法を開発した。
2.新しいレセプターアッセイ法を開発し、デングウイルス感受性ヒト赤芽球系細胞に、デング2型ウイルスと特異的結合性を持つタンパク質および糖脂質の存在を明らかにした。
東南アジアで流行しているデング2型ウイルスが吸着・感染増殖できる標的細胞としてヒト赤芽球由来の細胞(K-562)を上記の方法により特定した。今回、この細胞を大量培養し、そこから、デング2型ウイルス(D2T7)と特異的結合性を示す物質を探索した。その結果、デング2型ウイルスと特異的に結合するタンパク質(おそらく糖タンパク質)および糖脂質の存在を明らかにした。ウイルス結合性タンパク質はSDS-PAGEによってタンパク質を変性させた状態でもウイルスと結合性を示したことから糖タンパク質であり、ウイルスはそれに含まれる糖鎖へ結合している可能性が強く示唆された。また、本タンパク質は、宿主細胞膜ミクロドメイン(Lipid Raft)に存在する可能性が示唆された。
3.ヒト赤芽球系細胞からデング2型ウイルスと特異的に結合する糖脂質の単離に成功した。
ヒト赤芽球系細胞(K-562)細胞をクロロフォルム/メタノー抽出、アルカリ水解、イオン交換および吸着クロマトグラフィーなどにより純粋な形で分離することができた。本糖脂質は、TLC上でGM1aとGD1aの間に展開される。現在、この化学構造を解析中である。また、この糖脂質に対するモノクローナル抗体を作成している。これにより、本物質の宿主動物組織・細胞における局在性が明らかにできる。
