ナトリウム利尿ペプチドは魚類の広塩性を支配するか:トランスジェニック法による解析

研究概要

本年度は、ナトリウム利尿ペプチド(NP)ファミリーの分子進化と、新しい浸透圧調節ホルモンである可能性を持つアドレノメデュリンファミリーの発見で大きな発展があった。
1)NPファミリーは、これまで魚類ではANP, VNP, CNPの3種がファミリーを形成していると考えられてきたが、私たちはチョウザメでANP, BNP, VNP, CNPが共存することを明らかにした。その結果、ほ乳類で知られていたBNPと硬骨魚類の真骨類で知られていたVNPが別物であることがわかった。そこで真骨類でBNPを探したところ、ティラピア、メダカ、フグ、キリフィッシユ、ヒラメなどでBNPを見つけることができた。また、真骨類には4種のCNPが存在し、ほ乳類のCNPはCNP-4であり、CNP-3からANPとBNPがタンデムに遺伝子重複により作られたことを明らかにした。いっぽう、もつとも原始的な脊椎動物である円口類に属するヤツメウナギには、CNP-4のみが存在することが明らかになった。したがって、NPファミリーの祖先分子はCNP-4であり、硬骨魚類へと進化する過程で大規模な多様化が起こった可能性が高い。
2)ほ乳類で多機能ホルモンとして知られているアドレノメデュリン(AM)が、フグでは5つのパラログからなるAMファミリーを作っていることを明らかにした。5種のうち、フグAM-1がほ乳類AMのオルソログであることがリンケージ解析により明らかになった。いっぽう、AM-1/4/5とAM-2/3が異なるグループに属することがエキソン・イントロン解析や系統解析で示唆されたため、ほ乳類でAM-2を探したところ、ヒト、マウス、ラットでAM-2を発見することができた。AM-2は強力な心血管作用や腎作用を持つため、血圧や体液を調節する主要なホルモンであると考えられる。
