テンサイにおける細胞質雄性不稔の原因遺伝子の同定と花粉退化メカニズムの解明

研究概要	今年度得られた研究成果は次の通りである。 [1]Owen細胞質雄性不稔株ミトコンドリアゲノムの全塩基配列 Owen細胞質のミトコンドリアゲノムの全一次構造を決めた。ゲノムサイズは501020塩基対であり、G+C含量は43.9%と計算された。得られた塩基配列の7.7%はテンサイ正常株配列も含めて、いかなる配列とも相同性を示さない。 [2]Owen株におけるミトコンドリア遺伝子の変異 Owen株ミトコンドリアゲノムは正常株ミトコンドリアのコードする51種の遺伝子すべてを含んでいる。遺伝子コード域をOwen株と正常株間で比較した結果、tRNAとrRNAコード域には全く変異が見当たらなかった。一方、Owen株のタンパク質コード遺伝子については24箇所の点突然変異が見出された。アミノ酸配列の変化を伴う変異も含まれているが、進化的保存サイトではなかったり、あるいは同一のアミノ酸残基をもつ植物種が報告されているなどの理由により、これらの点突然変異が遺伝子機能を損なう可能性は極めて低いと考えられる。 [3]Owen株固有配列の発現解析 Owen細胞質雄性不稔株固有のORFやキメラ遺伝子の発現を調べた。これら固有配列をプローブに供してノーザン解析を行ったところ、4遺伝子に関して明瞭なシグナルバンドを検出した。稔性回復核遺伝子との相互作用を調査した結果、いずれにおいてもシグナルパターンの変化は見られなかった。現在それぞれの翻訳産物に対する抗体を作製し、ウエスタン解析を進めている。
