研究概要

カイコ幼虫中腸上皮細胞のアピカル膜を調製してバシラス・スリンジェンシスの生産する殺虫トキシンとの相互作用を研究した。主要な4大成果を得た。1)Cry1Aa,Cry1Ab,Cry1Acと結合する、新規結合性蛋白質P252を発見した。2)Cry1Acとだけ結合性する、カテゴリー1に属する新規アミノペブチデースP96を発見した。3)アピカル膜蛋白を組み込んだリポソームは、含まないリポソームの2倍の結合能を3種のCry1Aトキシンに対して示した。4)Cry1Aaはアピカル膜に結合した後一部を膜に陥入すると考えられるが、陥入部位が24kDaで、α-ヘリックス2からα-ヘリックス7の部位である事が初めて示唆された。
P252はTritonX-100,CHAPS可溶性画分に回収されラフト以外の部位に存在する事がわかった。サホモ4量体のサブユニット構造を持つと思われた。糖タンパクでありレクチン結合アッセイからバイセクティングGlcNAcを含む3本鎖のN-結合型糖鎖とGalNAcを含むO-結合型糖鎖を持つと考えられた。3種Cry1Aとの結合定数はそれぞれ、10nM,70nM,20nMで文献的に最も強い結合の一つであった。異種競合アッセイの結果は、P252のCry1Aa結合サイトはCry1Acと50%シェアし、Cry1Ac結合サイトは80%Cry1Aaとシェアリングした。Cry1Abの結合定数は70nMで、生理的解離結合と考えられたが興味ある事に、Cly1Aa,Cry1Acと90%ほど共有し、BSAによっても阻害された。
P96はアミノペブチデース1の抗体によって認識された。Cry1Acだけと結合し、結合はGalNAcで阻害された。基質特異性から中性アミノペプチデースと判断した。反応は2価の金属イオンを要求し、最適pHは6〜8であった。基質分解反応は2相を示し、高濃度基質で反応が促進された。
