研究概要	1)この2年問は、われわれが過去15年間の間にオオムギからクローニングしたムギネ酸生合成系路上の遺伝子群のうち、Ids2(Iron deficiency specific clone no.2)とIds3(Iron deficiency specific clone no.3)に関して、その上流解析を行ってきた。すなわち、両遺伝子は鉄欠乏のオオムギ根で特異的に発現する。そこで、これらの遺伝子はまだこれまで植物で解明されていない「鉄欠乏応答性シスエレメント」を有しているはずであるので、その同定作業に取り掛かった。約2kbのIds2promoterを大まかに分断し、それぞれの断片のpromoter-GUSをタバコに導入し、まず500base上流に最小の鉄欠乏応答性シス領域があることを見出した。この500baseの領域をさらに3'側と、5'側から削った各数種類のpromoter-GUSのdeletion seriesを作成し、タバコに導入し、40残基ずつの二箇所のシス領域を確定した。さらに、このうちのひとつの領域に関して、この40残基に3残基ずつ変異を導入したdeletion seriesを作成し、ほぼ19残基の有効な鉄欠乏応答性シスエレメントを確定した。現在、もう一方の40残基の領域に関しても、同様な手法で領域を狭めて有効な鉄欠乏応答性シスエレメントを確定する努力をしている。2)オオムギのゲノムのニコチアナミン合成酵素遺伝子(NAS)とオオムギのゲノムのニコチアナミンアミノ基転移酵素遺伝子(NAAT)をタンデムに連結して、イネに導入して、アルカリ土壌を用いてポットでの鉄欠乏耐性を検定した。2世代検定し有望な株を選抜した。
