NKT細胞の分化と機能維持の分子メカニズム

研究概要

NKT細胞による細胞傷害の分子機構:昨年、NK細胞やNKT細胞の機能を制御する新規C-タイプレクチンに属する"NKG2I"を新規同定した。この機能をより明確に示すためにhomologous recombinationの技法を用いてNKG2I遺伝子欠損マウスを作成した。NKG2I遺伝子欠損マウスにおけるNK細胞、NKT細胞の分化成熟は正常であったし、個体の発育に関しては全く以上を認めなかった。NKG2I欠損マウスのNK細胞のアロ標的細胞に対する細胞障害活性を失い、放射線照射した欠損マウスに移植したアロ骨髄細胞の生着を促進した。以上の結果から、NKG2Iは新規の活性化型NK受容体であり、アロ骨髄移植に関与していると考えられた。
NKT細胞機能の分子基盤:NKT細胞の特徴的正常が明らかとなった。それは、NKT細胞は特異的リガンド刺激によって、通常のT細胞に比べてアポトーシスに抵抗性であり、それはMyD118,NAIP2などのアポトーシス抵抗遺伝子の過剰発現がおこることによることが判明した。しかも刺激直後は受容体の発現低下を来すことがわかった。また、通常にT細胞では抗原刺激後の受容体発現低下の後,免疫寛容に陥り、機能不全になるが、NKT細胞の場合は寛容に陥らず、サイトカイン産生は正常に機能していた。これらの性質はNKT細胞の免疫制御機能を発現する上で重要な性状であると考えられた。
NKT細胞の自然免疫系における役割:NKT細胞の免疫システムにおける役割が明らかとなった。それは、NKT細胞が、自然免疫系と獲得免疫系を橋渡しする細胞であることが証明されたからである。NKT細胞は内在性抗原とCD1d分子により絶えず弱く活性化されており、従ってIL-12受容体、CD44、CD69の発現を示す。しかし、この状態ではサイトカイン産生まで至らず、機能しない。しかし一度細菌感染などでIL-12産生が引き起こされるとIL-12からのシグナルがNKT細胞を強力に活性化しIFNγなどのサイトカインによって、自然免疫系と獲得免疫系野細胞を活性化する。このような理由から、NKT細胞は素早く反応する細胞であるとともに、自然免疫系と獲得免疫系を橋渡しする細胞といえる。
