骨髄幹細胞の可塑性と多様性の解析

研究概要

1)昨年度までの研究では肝臓、神経、筋肉は勿論のこと、造血幹細胞が血管や血管内皮に分化することも確認できなかった。これはhemangioblastの概念や、最近話題になっている血管内皮前駆細胞(EPC)の存在、骨髄細胞の可塑性を期待した血管新生を促す臨床治験が行われること、などを考えると納得のいかない結果である。そこで、今年度はGFP陽性全骨髄細胞を移植したマウスの大体動脈をligationして、その後の血管新生の様子を解析した。結果はやはりネガティブで、GFP陽性細胞による血管新生は認められず、Ligationモデルにおいてもやはり骨髄細胞由来の関与は認められなかった。これらの結果は報告されている骨髄細胞の可塑性と相容れない結果であり、臨床治験を支持する基礎データにはなっていない。
2)骨髄細胞中に含まれていると報告されているEPCの存在を確認することを目的として、血管内皮前駆細胞を同定するアッセイ系の確立を試みた。ストローマ細胞(OP9) monolayer上に骨髄細胞を撒き、VEGF, FGF, EGFの存在下で培養すると10日程度でVE-cadherin陽性(血管内皮)細胞より構成されるコロニーが形成される条件を見出した。さらにこういったコロニーを形成する細胞のフェノタイプを決めるためFACSやMACSによる細胞分離と分離した細胞によるコロニー形成能を解析したところ、コロニー形成能を有する細胞はSca-1陽性分画細胞中に認められた。
3)lnkというアダプター分子を欠くlnkKOマウスでは造血幹細胞数が約16倍程度に増加していること、造血幹細胞の数だけでなく個々の幹細胞の自己複製能も増強していることを明らかにした。これらの結果はlnkが造血幹細胞の自己複製能と強く関連していることを示唆しており、自己複製に関与するシグナル機構を解明するに貴重なてがかりとなると考えられる。また、lnkが造血幹細胞の数のみならず自己複製能を増強するということはlnkの発現を低下させれば造血幹細胞を標的とした遺伝子治療や細胞療法の際に選択的有意性を与えることができると予想され、siRNAやdominant negative lnkを作成することが有用と考えられる。
