腎臓チャネル病の細胞内病態に基づく治療戦略の研究

研究概要

1.AQP2に関しては、優性遺伝型式の発症が認められている3家系の腎性尿崩症恵者の遺伝子解析を行ない、いずれもC末側のフレームシフト変異であることを見つけたこの変異蛋白と正常AQP2蛋白が4量体を形成することを明らかにした。変異蛋白はアフリカツメガエル卵の発現系では細胞内に留まり細胞表面には出現しなかった。
2.培養MDCK細胞を用いて、AQP2をstableに発現する実験系を確立した。通過性のある膜上での培養で、cAMPの刺激によってAQP2が細胞内小胞から管腔膜へ移行することを共焦点顕微鏡ならびに免疫電顕で確認することができた。上記の優性遺伝のAQP2変異を同じ手法で発現させたところ、この変異AQP2蛋白は膜上に発現させた場合にのみ、側底膜に局在しておりcAMPの刺激にも無反応であった。現在変異のどの部分がこの局在の変化に関与しているかを種々の変異蛋白の発現実験から絞り込んでいる。またこの現象をin vivoで再現するためにこの変異遺伝子のトランスジェニックマウスを作成中である。
3.CLCクロライドチャネルについては、CLC-3のノックアウトマウスの作成に成功したので、その形質の確認とメカニズムについて検討を行なった。ノックアウトマウスは多動、発育不全、視力障害を示し、組織学的検討では腸管上皮の脱落、海馬・網膜の萎縮を認めた。
4.記のCLC-3ノックアウトマウスの症状はヒトneuronal ceroid lipofuscinosisに近似しており、ライソソーム内にミトコンドリアF1F0ATPaseサブユニットCが蓄積していることも確認できた。これらのデーターはCLC-3が細胞内小胞のクロライドチヤネルとして働いていることを示しており、さらに詳細な機構の解明を行なっている。
