研究概要

我々のグループは胃・腸を中心に消化管の生理機能解明、とりわけ、胃酸分泌を担う胃プロトンポンプや腸における慢性下痢疾患や薬物による下痢疾患に関わる塩素イオンチャネルについて研究を展開してきた。我々は胃酸分泌細胞から得られた胃ベシクルの2分子膜の外層(細胞質側)より内層(細胞外側)に、ATP依存的にPC, PS, PEといった広範囲なリン脂質を輸送するフリッパーゼを発見した。本年度は胃フリッパーゼについて以下のような究を行った。SCH28080のアフィニティラベル物質を有機合成し、この特異的阻害剤が結合したタンパクを精製する実験を行ってきた。胃フリッパーゼが胃プロトンポンプと共存し、胃プロトンポンプの阻害剤により、胃フリッパーゼの機能も阻害されることから、両者は同一の分子の多機能である可能性が強い。また、分子がプロトンポンプ機能を行う際に、膜脂質を鰹削り器のように、削って、移動させるというコンピュターモデルを構築した。他方、胃酸分泌細胞膜の基底側膜に細胞防御に直接関わる塩素イオンチャネルが存在することを我々は発見した。このHousekeeping塩素イオンチャネルについて、このチャネルがこれまでに知られている塩素イオンチャネルfamilyのCLC-2, CLC-3, CLC-5である可能性について、チャネル蛋白質の抗体を作製して免疫組織染色により検討している。これまで、CLC-2は胃酸分泌細胞の分泌側膜に存在して、塩素イオン分泌に関与するという他研究者のこれまでの発表は間違っていることを発見した。
