核移植技術を用いたマウスES細胞由来個体の作出に関する研究

研究概要

最近、ES細胞を除核未受精卵へ核移植することによって生殖能力を持つマウスが得られるようになった、しかしながら、核移植卵の体外における胚盤胞への発生率は高いが、産子への発生率は1〜3%と低く、また得られた産子の多くは分娩直後に死亡する事が明らかになっている。本研究では、高率にES細胞由来の個体を得る事を目的として実施する。本年度は、核移植卵の発生能に及ぼすES細胞の遺伝的背景、核移植方法、細胞の細胞周期、発生刺激ES細胞の培養液などの違いの影響を検討した。近交系マウス並びに交雑系マウスから樹立した4株のES細胞を用いて核移植を行った結果、胚盤胞へはいずれの細胞株を用いた場合でも高率に発生した。受胚雌へ移植後、移植した核移植卵の内〜4%が産子に発生したが、発生率は細胞株間で大差なく、また得られた産子の多くは分娩直後に死亡した。細胞を除核卵細胞質に融合する方が、注入するよりも胚盤胞への発生率が高かったが、産子への発生率には大差なかった。また、M期の細胞を用いた場合、G1期の細胞を用いた場合よりも発生率が高かった。発生刺激法としてストロンチウム処置と電気刺激法を比較した結果、大差はなくいずれも産子へ発生した。これらの要因を検討する事によって、核移植卵の産子への発生率は1〜6%と若干向上したが、得られた個体の死亡率に改善は見られなかった。核移植卵と受精卵の蛋白質発現状況を2次元電気泳動を用いて比較検討した結果、違いの見られるスポットが複数個観察されたが、その詳細はさらに解析中である。
