核移植技術を用いたマウスES細胞由来個体の作出に関する研究

研究概要

本研究は、マウス胚性幹(ES)細胞由来のクローン個体を高率に作出できる核移植技術を確立する事を目的として実施した。本年度は、核移植卵の卵割時期が発生能に及ぼす影響について検討した。ノコダゾール処置によってM期に同調したTT2ES細胞を、除核未受精卵に不活化センダイウイルスを用いて融合した。ついで、ストロンチウム処置によって活性化刺激後、12時間目から22時間目まで1時間ごとに卵割時期を調べた。つぎに、卵割時期ごとに核移植卵を体外培養して胚盤胞への発生率を調べると共に、受胚雌に移植して胎子への発生率を調べた。その結果、体外受精卵と比べて、卵割時期のばらつきが大きい事が判明した。また、体外受精卵では早く卵割すればする程胚盤胞への発生率が高かったが、核移植卵では早く卵割した卵子や遅く卵割した卵子では中間区の卵子に比べて胚盤胞への発生率が低い事が判明した。また、胎子への発生率も中間区の卵子で高かった。次に、核移植卵におけるOct-3/4, Stat3, Nanog, Stelll遺伝子の発現量をリアルタイムPCR法を用いて、受精卵の場合と比較検討した。その結果、発生段階によって遺伝子発現量に差異が見られる事が判明した。また、核の初期化に関係すると考えられる因子を、牛卵子を用いた2次元電気泳動法によって解析した結果、初期化因子はこれまで考えられていたようなMPFやMAPKではない事が判明した。初期化に関連すると考えられる23kDの蛋白質を1個見い出し、塩基配列の結果からfortilinであることが判明した。
