TTウイルスの遺伝子型分類法の開発と感染病態解明への応用

研究概要	TTウイルス(TTV)というウイルス名は発見の発端となった患者のイニシャル(T.T.)に由来する暫定的な名称であるが、今年度、国際ウイルス命名委員会(International Committee for the Taxonomy of Viruses : ICTV)によって、正式名称が決定された。命名にあたって、環状の遺伝子構造にちなんで"ネックレス"や"指輪"の意味のラテン語が用いられ、ウイルス名は略称が同じくTTVとなるように「TorqueTeno virus(トークテノウイルス)」、ウイルス属は「Genus anellovirus(アネロウイルス属)」となった。ウイルス科については、新しいウイルス科を設定する方向で現在引き続き検討されている。このStudy Groupからの要請に応じ、これまでの研究成果を整理して提供した。それらがTTVの分類、特徴付けに大きく反映されている。今年度までの研究により、TTVは大きく5つのグループ(phylogenetic groups)に分類できることが分かった。そして、少なくとも40種類の遺伝子型に分類できることが明らかになったが、現時点でもTTVの多様性の全容解明には至っておらず、データの蓄積が必要であると考えている。5種類のグループに特異的なプライマーを設計し、selective PCR法によりグループ別の頻度を調査した結果、小児期に最も早く感染し、陽性頻度の高いTTVグループがグループ4であり、成人では感染者の約半数がこのグループ4のTTVに感染していることが分かった。一方、患者T.T.から最初に分離されたプロトタイプのTTV(遺伝子型1型)を含むグループ1のTTVは主として生後6ヶ月以降に感染し、成人での陽性率は約15%であった。現在、各TTVグループ別の感染率および感染状況を各種疾患別に検討中である。
