研究概要

1)先にTCDDが、Wntシグナル伝達系を介して脳神経系の発達毒性を発現する可能性を見出した。そこでTCDDへの周産期曝露による胎仔脳及び肝内遺伝子の発現変化を、DNAマイクロアレイ法で、より網羅的に解析し、発現が変化する遺伝子群を見出した(鏡)。また、周産期にTCDDへ曝露された成熟ラットにおける海馬依存性学習行動の変化を検討し、雌雄差が消失することを見出した。この変化には、海馬CA1領域のCREBの活性低下が関与する可能性が高い。さらにc-Srcは、TCDDの毒性発現への関与が示唆されているが、そのリン酸化部位Ser75をAspに変えたES細胞株を作製した(前田)。
2)AhR repressor(AhRR)::Nrf2 2重欠損マウスを作製し、ベンツピレンによる皮膚発癌実験を行った。AhRR単独欠損マウスは発癌に対し抵抗性を示すが、AhRR::Nrf2 2重欠損マウスは示さなかった。従って癌抵抗性は、Nrf2に依存すると考えられる(山本)。
3)野生型及びPPARα欠損マウスにDEHPを投与し、腫瘍発生臓器における遺伝子発現変化をDNAマイクロアレイ法で解析したところ、野生型の腫瘍発生部位においてはc-jun mRNA量が、PPARα欠損マウスの腫瘍発生部位にはc-jun、junD1、c-Myc mRNA量が著しく増加していた。従ってc-jun mRNA量の増加にはPPARαが関与しないと考えられる(那須)。
4)先にTCDD感受性の違いに関与する因子を同定するため、TCDDへの感受性が高いと考えられる家族性高TSH血症3家系の全ゲノムにわたる連鎖解析を行い、4番染色体短腕に、形質と連鎖する遺伝子座を見出した。そこで、4p16領域に存在するSolute Carrier FamilyであるSLC26A1およびSLC2A9の配列を決定したが、発症者に特異的な変異を見出さなかった(小泉)。
