研究概要	歯周病は成人における歯牙喪失の主因であり、国民の40%以上が罹患する、いわば国民病のひとつである。本疾患は歯周局所の常在微生物によって惹起される感染症で、とくにグラム陰性嫌気性細菌のジンジバリス菌は主要な原因菌とされている。本菌はArg-gingipain(Rgp)とLys-gingipain(Kgp)という2種類のシステインンプロテアーゼを産生し、宿主に対しては強力な病原性を発揮する破壊因子として、菌自身には歯周ポケットで本菌が発育・増殖するための生存因子として働いていることが示唆されてきた。本研究は、これら2種類の酵素に対する特異的な阻害剤を開発し、本菌の病原性を抑制し、歯周病の治療・予防に向けた新たな方法論を確立することを目的としている。昨年度の研究から、Kgpに対する特異的なペプチド性合成阻害剤を開発することができたが、本年度の研究ではRgpに対する特異的な天然ならびに合成の阻害剤が開発された。天然の阻害剤としては、放線菌の1種Streptomyces sp. Strain FA-70から分子量606でC_<27>H_<43>N_9O_7の分子式を有する化合物(FA-70-C1と命名)を単離した。この化合物は合成基質に対して4.5 x 10^<-9>Mの阻害定数を示した。この化合物は宿主のプロテアーゼであるカテプシン群に対しては弱い阻害活性しか示さなかった。この化合物は、ジンジバリス菌による好中球、線維芽細胞、ならびに血管内皮細胞の傷害活性を強く阻害した。また、in vivoにおける本菌によって誘発される血管透過性亢進を強く抑制した。一方、合成のペプチド性阻害剤としてリジン、アルギニンを基本骨格にもつトリペプチド誘導体の中からRgpに対する強い阻害活性をもつ化合物を十数種類合成した。そのうちの1つKYT-1はRgpを10^<-10>Mオーダーで強く阻害し、Kgpや他の宿主プロテアーゼを殆ど阻害しなかった。本化合物はジンジバリス菌によって誘導される好中球や線維芽細胞、ならびに血管内皮細胞の傷害活性を強く抑制した。以上の結果から、FA-70C1ならびにKYT-1がジンジバリス菌の病原性を阻害する有用な化合物であることが明らかとなった。
