研究概要	歯周病は歯周局所の常在微生物によって惹起されるある種の感染症であるが、なかでもグラム陰線嫌気性細菌Porphyromonas gingivalisは成人性歯周炎の主要な原因菌としてその発症進展に深く関わっている。これまでの研究から、本菌が産生する2種類の強力なシステインプロテアーゼRgpおよびKgpが協調的に働きながら、宿主には強い病原性を、菌自体には発育増殖のための必須の生理機能を発揮していることが明らかにされている。本年度は、両酵素の大部分を占める菌体結合型高分子複合体の分離精製とその性状を明らかにすることを第一の目的とした。野生株(ATCC33277)を非イオン性界面活性剤で処理した後、遠心分離で得られる上清をDEAE-Sephase1およびSepharose 6Bのカラムクロマトグラフィーにかけた。酵素活性画分をAnti-Rgp antibody-Sepharoseのアフィニティーカラムにかけた後、活性画分をショ糖密度勾配遠心にかけた。得られた最終酵素標品は未変性のゲル電気泳動で単一のバンドを示した。この分子はRgpおよびKgpの酵素活性ドメインとそれぞれのC末端付着サブドメインをもち、さらにリン脂質およびLPSを含む600kDa以上の膜結合型高分子複合体であった。この複合体の酵素学的性質は、酵素ドメインのみから成るそれぞれの単量体分子の性質と類似していたが、コラーゲン分子に対する分解活性は複合体分子の方が単量体分子の総和よりも明らかに強かった。また、ヒト歯肉線維芽細胞や血管内皮細胞に対する傷害活性も複合体分子の方が単量体分子よりも強かった。以上の結果は、RgpおよびKgpは菌体外膜上で会合し、リン脂質やLPSを結合して高分子ヘテロ複合体を形成し、より安定で強力な酵素活性をもつ病原性分子として重要な機能を果していることを強く示唆している。
