自己幹細胞を用いた自己組織再生誘導型バイオ人工臓器

研究概要	コラーゲンなどの天然高分子材料を用いて3次元細胞外マトリックス材料を作製し、動物実験により細胞外マトリックスという場において生体細胞群がどのような再生経過をたどるのか、細胞外マトリックスの性状と組織再生能とどのような関係をとるのかを明らかにすると共に、自己幹細胞を用いて自己組織や自己臓器を再生誘導するという全く新しいバイオ人工臓器を開発した。具体的には自己組織再生誘導式の人工血管、人工神経、人工食道、人工気管、人工気管支、人工小腸、人工胃、人工大腸、人工胆管、人工尿管、人工硬膜、人工心膜、人工膀胱、人工角膜、人工喉頭、人工声帯など比較的単純な臓器から、人工心臓、人工肺、人工腎臓、人工肝臓、人工膵臓などの多機能の複雑な実質的臓器までの再生誘導を動物実験的に検討した。体内に再生の足場となるコラーゲンを置くと、そこに生体由来の細胞が侵入して、その場で新しいコラーゲンを産生し、自己の組織を作り出して行く。生体はその再生を促進するための多種の因子をその場に放出して、その再生が進んで行くことが確かめられた。人間における臨床応用を念頭において、実験には大型動物を用いた。全く新しい臓器再建法を創出した。報告書末の文献リストに掲げたように、人体全体におけるほとんど全ての臓器組織でin situ Tissue Engineeringで再生が可能であることが判明した。引き続き、臨床応用を目指して研究を遂行して行く方針である。
