研究概要

1.シロイヌナズナ突然変異体を用いた解析:マッピングベースのクローニングを行い、van突然変異体のうち2つの変異遺伝子の候補を見いだした。
2.ヒャクニチソウ葉肉細胞からの管状要素分化系を用いた解析:
(1)ファイトスルフォカイン(PSK);管状要素分化誘導因子であるPSKの遺伝子をヒャクニチソウから単離し、その遺伝子産物の局在をGFPとの融合遺伝子により調べたところ、ゴルジ体に存在することが明らかになった。
(2)ブラシノステロイド;ブラシノステロイド合成の鍵酵素遺伝子CPDとDWF4のヒャクニチソウでのin situ hybrudizationを行った。比較的広い範囲での発現が見られた。
(3)細胞外分泌分化関連因子;新たな細胞間相互作用関連因子を単離するために、培地成分に対するファージディスプレイサブトラクション法による抗体ライブラリー作成を行った。その結果、多種のクローンを含むライブラリーの作成に成功した。
3.篩部制御遺伝子の単離と機能解析:維管束は木部と篩部のバランスの取れた形成により、機能的なユニットとなる。そこで、未成熟な篩部特異的発現を示すHD-Zip型のホメオボックス遺伝子ZeHB3を単離し、ZeHB3のシロイヌナズナでのDEX誘導型過剰発現体を用いてジーンチップ解析を行い、ZeHB3により再現性よく誘導される数種の遺伝子を単離した。これらの遺伝子の多くは篩部で特異的に発現していた。したがって、これらの遺伝子は、ZeHB3タイプのホメオボツクスの下流にある遺伝子である可能性が示された。
