研究概要

本研究では、ヒト抗体ゲノム情報に基づき、(1)遺伝子レベルの多様性の創出、(2)抗体の分子認識能による安定な生体外選択、(3)選択された抗体の高効率な調製系を構築することにより、生体内の免疫系を凌駕するような新機能ヒト抗体分子調製系の構築を目標としている。また、研究代表者らは、最近、大腸菌で大量に発現する不溶性顆粒から、抗体断片の高効率再生系を確立しており、選択された有用ヒト抗体断片はこの系を用いて調製し、治療・臨床検査での利用を目指している。本年度の研究業績は以下の通りである。
(1)遺伝子多様性創出:抗体可変領域への無作為変異導入法として、Error-pronePCR法をとりあげ、無作為変異導入が可能な条件を検討し、金属イオンと反応時間の調節が重要であることを見出した。これを用いて、ヒト型化した抗体へ無作為変異を導入、選択を試みることにより、数箇所への変異導入により親和性の顕著な上昇を見ることができた.
(2)前年度に研究(1)で構築された遺伝子ライブラリーをファージライブラリーとして、前年度と同様の選択を行った。標的抗原として、新規サイトカインならびに自然免疫関連受容体を取り上げたほか,自己抗原に加え生体に投与不可能な物質(例えば毒性物質や工学材料)を取り上げた。
(3)選択された抗体の高効率な調製系の構築と構造解析:(2)で得られたヒト抗体可変領域のうち、生分解性プラスチック特異的抗体可変領域断片について、大腸菌の高発現系で可溶性分子として調製し、QCMにより抗原認識能を調べたほか、その構造解析に相応しい結晶を得た。アルカリホスファターゼなどの酵素との融合あるいは2種類の抗体断片や免疫賦括剤(サイトカイン)との融合を試み、安定な調製系を構築した。モデル抗体の分子認識能を詳細に解析した。抗体調製に関する巻き戻しシステムの分光学的解析を行った。
