研究概要

本研究では、ヒト抗体ゲノム情報に基づき、(1)遺伝子レベルの多様性の創出、(2)抗体の分子認識能による安定な生体外選択、(3)選択された抗体の高効率な調製系を構築することにより、生体内の免疫系を凌駕するような新機能ヒト抗体分子調製系の構築を目標としている。また、研究代表者らは、最近、大腸菌で大量に発現する不溶性顆粒から、抗体断片の高効率再生系を確立しており、選択された有用ヒト抗体断片はこの系を用いて調製し、治療・臨床検査での利用を目指している。本年度の研究業績は以下の通りである。
1.選択:前年度までに研究(1)で構築した遺伝子ライブラリーをファージライブラリーとして、選択を行い、新たにNK細胞阻害性受容体(KIR)特異的ヒト抗体、金表面特異的抗体の選択・調製に成功した。また、BIACORE2000のフロー・リアルタイムモニターリングシステムを用いた、選択・調製法を構築した。
2.調製系:選択された抗体の高効率な調製系の構築と構造解析:(2)で得られたヒト抗体可変領域について、大腸菌の高発現系で可溶性分子として調製し、精密な結合定数を測定した。また、分子認識能の詳細な記述のために、前年度調製した生分解性プラスチック表面特異的抗体の結晶構造を明らかにし、フラットな面と疎水・荷電残基の局在化による特異的な認識機構を持つ可能性が強く示唆された。アルカリホスファターゼなどの酵素との融合あるいは2種類の抗体断片や免疫賦括剤(サイトカイン)との融合を試み、安定な調製系を構築した。抗体・各種サイトカイン等の調製に関して、添加剤の効果を考慮に入れた巻き戻し法の改良を試みた。
