昆虫細胞のカロチノイド透過性を支配する分子機構の解明

研究概要	昆虫体液リポホリンを精製すると黄色く見える。リポホリンは多くの脂質とともにカロチノイドを結合しているために黄色くなっている。植物由来のカロチノイドがどのような仕組みで中腸から体液や絹糸腺まで届けられているのかについてはまったくわかっていない。細胞内にカロチノイドに特異的に結合するタンパクがあって、移行に関わっているであろう事は長い間想像されてきたが、その実体は知られていなかった。本研究において、カロチノイド結合タンパク(CBP)を精製し、機能を解明した。このタンパクは、33kDaでルテインを1分子結合していた。CBPは絹糸腺、中腸と生殖腺に存在するタンパクであり、これらの組織はいずれも黄色に呈している組織であった。しかも、CBPはY遺伝子(黄色の体液)をもつ系統にのみ発現し、+^Y遺伝子(Yの劣性遺伝子で無色の体液)を持つカイコでは発現していなかった。CBPのcDNA解析から、CBPはSteroidogenic acute regulatory(StAR)と呼ばれるファミリーに属し、脂質結合部位にはコレステロールが結合するが、CBPはコレステロールの代わりにカロチノイドを結合する新たなStARであることがわかった。カロチノイドはリポホリンに結合して昆虫の体液中を移動する。カロチノイドが、絹糸腺など特定の組織に受け渡しされるが、選択的に特定の組織に移行しているメカニズムについては明らかにはされていない。しかしながらCBPが存在する組織のみにカロチノイドが積み下ろされており、このタンパクが細胞内のカロチノイド輸送やリポホリンからの取り込みに関わっていることと思われる。またリポホリンが特定の組織の細胞膜に結合するためのリポホリンドッキングタンパクや、リポホリンから脂質を取り込むためのLipid Transfer Particle(LTP)の存在を明らかにした。
