糸状菌糖質分解酵素遺伝子群の転写因子に関する機能ドメイン解析

研究概要	麹菌デンプン分解酵素遺伝子群、キシラン分解酵素遺伝子群の発現誘導は、転写因子AmyR、XlnRによりそれぞれ支配されている。また、麹菌アミラーゼ遺伝子では広域転写促進因子であるHap complexが転写促進に働いている。本研究ではこれら3種転写因子の機能解析を行った。 1.AmyRの機能解析AmyRは酵母Mal activatorと5ケ所の相同領域を共有している。これらの相同領域をC末側から順次欠失させ、AmyRのDNA結合能、転写活性化能、転写誘導能に与える影響を解析した結果、N末にDNA結合領域、中央部に転写活性化領域、C末に転写活性化の制御領域を持つことが明らかとなった。 2.XlnRの機能解析麹菌XlnR遺伝子破壊株ならびにXlnR高生産株を用い、XlnRの支配下にある遺伝子の同定を行った。その結果、キシラン分解酵素遺伝子群だけでなく、複数のセルラーゼ遺伝子も本転写因子の支配下にあることが明らかとなった。 3.Hap complexの機能解析麹菌Hap complexはAoHapB、C、Eの3種のサブユニットを含む。各サブユニットの部分欠失がDNA結合、転写促進に与える影響を解析し、各サブユニットのコア領域のみの複合体でDNA結合活性を有すること、AoHapBのC末側領域、及び、AoHapCのN末側領域に、転写促進機能保持に重要な領域が存在することを明らかにした。
