研究概要

萎凋枯死機構については、病原菌Raffaelea quercivoraが引き起こす通道阻害機構を明らかにするために、ミズナラ苗木の接種部付近の水分の変化を中性子ラジオグラフィで観察したところ、接種部のごく近傍では水分量が少なかったが、通道阻害域は乾燥域よりもはるかに大きく、比較に用いたスギとは異なり、本病における通道阻害では急激なキャビテーションは発生していない可能性が示唆された。病原菌接種によって通道阻害の引き起こされたミズナラ苗の道管を軸方向に連続的に解剖観察したところ、通道阻害要因としては通道阻害域の道管にはチロースが、仮道管領域には閉塞物質がみられた。通道域の道管がチロースで閉塞している場合の多くは下部で通道阻害域と接するか、阻害域を通過している道管であった。
防御反応に関しては、反応障壁においてリグニン様物質が早く木繊維を閉塞し防御反応としての重要性が示唆されたことからこの物質の正体を探った。防御反応の中心である反応障壁では総リグニン量が増加し、増加の大半は縮合型リグニンであった。非縮合型リグニンの中では傷害リグニンとして知られるグアヤシル型の割合が高かった。これらより、病原菌の侵入に対し通常のリグニンに比べてより強固なリグニンを作り出すことで防御壁を作っていると考えられた。
感受性および抵抗性樹種の反応特性解明では、病原菌の接種によって形成された軸方向と水平方向への材変色を樹種間で比較するため、接種ブナ、ミズナラ、コナラ、アラカシ、スダジイ、マテバシイ間の感受性の差異に関与する要因を明らかにすることを目的とした。軸方向の材変色長は個体差が大きく、樹種間の感受性の差異と一致しなかった。一方水平方向への材変色域の割合は、樹種間で有意な差がみられ、樹種間の感受性の差異とほぼ一致した。水平方向への材変色の拡大が樹木の萎凋症状に関与している可能性が高いと考えられる。
