魚類のC3レセプタータイプ2(CR2)の構造と機能:補体による獲得免疫の制御機構

研究概要	本研究は、硬骨魚類が哺乳類のCR2に相当するレセプターをもち、C3d-CR2の相互作用を介した免疫応答の制御機構を備えているかどうかを明らかにすることを目的とする。まず、CR2のリガンドであるC3dがコイでも生成するかどうかを調べた結果、異物表面上および血清中で補体活性化後6時間をピークとして精製することが確認された。次に、組換えC3dフラグメントを作成した。すなわち、C3d領域をコードするcDNA断片をPCR増幅し、これをpGEX-4T-1ベクターに連結し、コイC3dをグルタチオン-S-トランスフェラーゼとの融合タンパク質として発現させた。同時に、GSTの下流にC3dを2つ連結した融合タンパク質も作成した。これらの融合タンパク質およびGSTのみをコイの腹腔内に投与し、2週間後にコイ血中の抗GST抗体量をELISA法によって測定したところ、GST-C3d投与区の魚で、GST単独投与区の魚よりも若干高い抗体価が認められた。このことは、抗原にC3dが連結されていると、抗体産生が増強されたことを示唆している。すなわち、C3dとCR2との相互作用がコイの免疫応答に正の影響を及ぼしたのかも知れない。現在、コイリンパ球とGST-C3d融合タンパク質との相互作用を解析中である。一方、哺乳類のCR2のアミノ酸配列を用いて、フグゲノムデータベースを検索したところ、数種の相同な遺伝子の候補が見つかった。現在、この情報をもとにPCRプライマーを設計し、コイ白血球ライブラリーからCR2様のcDNAの増幅を試みている。
