乾燥・高温環境に適用できる植物成長制御法と新植物の開発に関する研究

研究概要	乾燥・高温環境における植物の定着に重要な芽ばえの伸長成長および根の姿勢制御、さらに高温ストレスによる生殖障害について、それらの制御と発現にかかわる分子機構を究明するための解析を行った。乾燥地における芽ばえの定着に重要であることがわかった深播き耐性は、第一節間のジベレリン超感受性によってもたらされ、それに伴うカリウムイオンの取り込みと「ねじれ」成長によって出芽を可能にすることを見出し、現在、その制御分子を同定する研究を進めている。シロイヌナズナの根の水分屈性誘導系を開発し、水分屈性におけるアブシジン酸とオーキシンの関与、水分屈性と波形成長の関係を明らかにした。また、水分勾配が重力センサーのデンプン-アミロプラストを速やかに消化して重力応答性を低下させ、それによって水分屈性が重力屈性に打ち勝って発現する機構を発見した。さらに、シロイヌンズナの水分屈性突然変異体を初めて単離し、rhy(root hydrotropism)と命名した。その中でrhy1は水分屈性をまったく発現しないが、伸長成長、重力屈性、光屈性、波形成長が正常な、水分屈性特異的突然変異体であった。一方、rhy2は、重力屈性、光屈性、伸長成長は正常であるが、水分屈性と波形成長が低下した突然変異体であった。rhy3は、光屈性と波形成長は正常であるが、水分屈性、重力屈性、伸長成長の低下した突然変異体であった。現在、これらの突然変異原因遺伝子のマッピングを試みており、rhy1の突然変異遺伝子は第1染色体と第4染色体には座乗しないことがわかった。このように、乾燥地への適応に重要な水分屈性の発現機構を分子レベルで解明することが可能になった。オオムギで雄性不稔を誘発する高温障害の実験系を開発し、幼穂形成初期に昼温30℃・夜温25℃を5日間処理することによって、その後の花粉形成が阻害され、完全に不稔になることを見出した。また、SAGE(Serial Analysis of Gene Expression)法およびNorthern法によって、高温によって特異的に発現が影響される遺伝子を見出した。この不稔は、胞原細胞およびタペータム細胞の高温障害に起因することが明らかになり、それらの発達初期に上昇するヒストンH3およびヒストンH4遺伝子ファミリーに属する遺伝子の発現が高温処理によって抑制されること、また60Sリボゾームタンパク質をコードする遺伝子の発現が高温処理によって低下することが見出された。さらに、この場合の高温処理は胞原細胞およびタペータム細胞の発育に特異的に作用し、栄養成長器官の発育に影響を及ぼさないことが明らかになった。
