抗SRYリボザイム及びアンチセンスSRYトランスジーンによる家畜の性比制御の試み

研究概要	本研究の目的は、哺乳類の性決定遺伝子SRYの転写産物に対して、アンチセンスmRNAあるいは、抗SRYリボザイムを遺伝子操作による導入遺伝子として発現させ、出生時性比が雌に傾斜するような、人工的家畜品種を作出するための基礎・開発研究を実施することにある。この目的を達成するために、まず、マウスをモデルとし、現在、最も強力なpromoter/enhancer complexをもつ哺乳類細胞における発現ベクターであるpCAGGS(阪大宮崎-純教授から供与)を用いて、アンチセンスSryを発現させたトランスジェニックマウス(C3B6Fl)を作出し、その生殖巣、生殖器官における表現型の解析を行った。これまでに、12匹(雄10、雌2)のトランスジェニックマウスが得られたが、表現型と遺伝型は全て一致していた。これらのマウスをfounderとして、非トランスジェニックC3B6F1マウスと交配して、Flを得たところ表現型が雌でありながら、PCR解析でY染色体のマーカーであるZfyに対して陽性を示す個体が、39匹のトランスジェニック雌マウス中11匹得られた。現在、これらのマウスについて、なお詳細な表現型および遺伝的解析を行っている。一方、抗マウスSryリボザイムを、ハンマーヘッド型、tRNA-ribozyme complex型等、数種類を設計した。これらのリボザイムにつき、in vitroにおける、Sry mRNAに対する切断効果を検討した結果、切断効果をもつことを確認した。また、これらリボザイムをin vivoで発現するためのpCAGGS系ならびにpolIIIプロモーター系の発現ベクター(pPUR-KE、東大多比量和誠教授から供与)の構築も完了した。今後、これらのin vivoにおける効果を検討する計画である。
