研究概要

新規摂食抑制ホルモンであるニューロメジンU(NMU)のノックアウトマウスは行動が緩慢であり、行動リズムが消失していることが判明した。そこで、種々の反射機構を検討した結果、熱反射や痛み反射がNMUノックアウトマウスでは劣っていることが判明した。また、NMUを脳室内に投与するとこれらの反射機構は亢進した。NMUの免疫陽性細胞が脊髄に分布し、またNMUの投与により脊髄にcFosが発現した。NMUノックアウトマウスは光への同調も遅く、環境温度への適応も低下していた。以上のことから、NMUは種々の反射機構を調節するホルモンであると思われた。一方、NMUのレセプターmRNAが時計部位の視交叉上核に存在し、24時間のリズムを示していること、また、NMUの脳室内投与により、視交叉上核にもFosが発現すること、さらにNMUの免疫染色で、視交叉上核の外背側部で染まることから、NMUは視交叉上核内でオートクライン的作用により、時計を調節している可能性が示唆された。また、NMUの類似体として新たに同定したNMSは、体内時計の局在部位である視交叉上核(SCN)に限局して発現しており、脳室内投与によって、投与時刻依存的に、行動リズムの位相変位を起こした。また、位相前進が観察されたサーカディアン時間の6時(CT6)において、NMS投与後SCNでのc-Fos蛋白質の発現および多ニューロン発火活動頻度の上昇が観察された。以上の結果、NMSはSCNの時計機構そのものの構成物質ではなく、時計を調節する物質であることが示唆された。
