ロタウイルス非構造蛋白のエンテロトキシン分子の特定とその下痢防御への利用

研究概要

本研究の目的は、ヒトや様々な動物に重篤な下痢を引き起こすロタウイルス感染症の発病機構を、ロタウイルス非構造蛋白質NSP4に焦点を絞り、分子生物学的に解明すると共に特定した下痢原性分子あるいはNSP4に対するモノクローナル抗体(MAb)を下痢防御へ応用することである。本研究では当研究室において世界で初めてハトから分離したトリロタウイルス、PO-13株を主に用いた。PO-13株は各種生物および遺伝性状が明らかになっていること、そのNSP4のアミノ酸配列が他のロタウイルス株のそれと大きく異なっていることにより、ロタウイルス共通のエンテロトキシン分子を容易に特定できると考えたからである。研究初年度は、PO-13株が哺乳マウスに下痢を起こすことを明らかにし、そのNSP4のアミノ酸109-135位の領域がエンテロトキシン分子であることを特定した(Virology,2001;J.Virol.,2002;Virus Res.,2002)。2年目は、主にNSP4に刻するMAbを作製し、それらによるNSP4の抗原性状を明らかにした(Microbiol. & Immunol.,2003)。そこで、最終年度の本年は、NSP4以外の下痢発現機構の有無、下痢症を起こす細菌で誘導される酸化窒素(NO)あるいはその前駆体であるiNOS蛋白質やmRNAが、NSP4やロタウイルス感染で誘導されるか否か、また、MAbがiNOSの誘導を抑制するかを検討した。その結果は以下の成果が得られた。
1.マウスに下痢を発現する株としない株間の分節遺伝子交換(リアソータント)ロタウイルスを作製して下痢発現の有無を検討したところ、NSP4とは別にウイルス外殻蛋白質の胃あるいは腸管での抵抗性が病原性に大きく関わっていることを明らかにした(Virology,2003)。
2.NSP4で感作あるいはウイルスで感染されたマクロファージ由来培養細胞、腸管マクロファージ細胞、摘出マウス回腸はNO,iNOS蛋白質あるいはmRNAを誘導し、下痢発現とNO誘導とは密接に関わっていることが示唆された(第51回日本ウイルス学会で発表、2003)。
以上の結果から、ロタウイルスの新たな下痢発現機構と防御作用の一部を解明することが出来た。この成績は、次世代型ロタウイルスワクチンの開発のための重要な基礎情報と言える。
