研究概要

Ess1/Pin1欠損酵母とPin1-KOマウスそレてPin1およびPar14の阻害剤をツールとして用いて、これらペプチジルプロリルイソメラーゼ(PPIase)の機能を明らかにしていくことを目的にして研究を行った。現在までに、3つの大きな成果をあげた。1)Ess1/Pin1欠損出芽酵母と野生型酵母の発現遺伝子をDNAマイクロアレイによって比較した。Ess1/Pin1欠損酵母ではパルブリン様PPIaseではないが、PPIaseグループに属するサイクロフィリンの発現が増加することを発見した。致死にならない原因はサイクロフィリンがEss1/Pin1の欠損を補って機能しているからだと推定できた。(Fujimori et al. Biochem Biophys Res Comm, (2001)289(1), 181-90)、この他、proteinチップでも同様に比較して検討する準備を行った(斉藤、内田(2002)癌の薬剤耐性診断の新展開臨床病理レビュー特集号(臨床病理刊行会 印刷中)、2)Pin1-KOマウスの表現型を詳細に検討して、cyclinD1-KOマウスと表現型が共通であることから、Pin1がcyclinD1の発現を制御している可能性を示した(Liou YC et al. Proc. Natl. Acad. Sci. USA(2002)99, 1335-1340),3)Pin1が細胞周期S期の進行のタイミングに関係することを、Pin1欠損胎児線維芽細胞を用いて示した(You et al, J Cell Biochem. (2002)84(2), 211-216)。また、新規な阻害剤を合成する生物化学的生合成法の準備を行った(Uchida et al(2002)Int J. Mol MEd, 9(2)141-145)。
