新規高分子量G蛋白質と結合分子群の機能協関および細胞内局在の動態

研究概要	マウス脳新規高分子量GTP結合タンパク質の機能解析を開始するにあたり、初年度は相互作用する蛋白質を同定することを目的に酵母two-hybrid法によるスクリーニングを行った。なお、本研究にて解析を行うマウス高分子量GTP結合タンパク質のヒトにおける相同分子が、遺伝性視神経萎縮の原因遺伝子であることが他のグループにより報告されOPA1と命名されたことより、以下マウスタンパク質をmOPA1と表記する。960アミノ酸よりなるmOPA1のほぼ全長に相当する部分(90-960の領域)をbaitとして用いて、マウス脳cDNAライブラリーを酵母two-hybrid法によってスクリーニングを行った。その結果、3量体Gタンパク質シグナル伝達系の調節因子であるRGSファミリーのサブタイプの一つであるRGS14、GABAA受容体サブユニットのサブタイプの1つである4サブユニット、mOPA1自身のC末端断片、および数種の新規遺伝子をmOPA1と相互作用する分子として同定することができた。欠失変異体を用いたtwo-hybrid法による検討により、mOPA1とRGS14間の相互作用はmOPA1のC末端付近に位置する645-885の領域と、RGS14の266-434の領域により行われていることが判明した。RGS14における相互作用する領域は3量体Gタンパク質のGTPaseを活性化するRGSドメインとは異なった領域であった。またmOPA1とGABAA受容体4サブユニット間においてはmOPA1の210-355の領域と、GABAA受容体4サブユニットのC末端付近において相互作用していることが判明した。次年度以降は神経細胞において、mOPA1タンパク質とこれら同定された結合分子群との細胞内分布と機能的意義をさらに解析していく計画である。
