糖鎖変異マウスを用いたスフィンゴ糖脂質の生体内機能の解明

研究概要	酸性スフィンゴ糖脂質の糖鎖が神経系の維持と再生において果たす役割について糖鎖遺伝子ノックアウト(KO)マウスを用いて検討した。これまで加齢に伴う末梢神経および中枢神経の変性が認められていたが、更に複合型ガングリオシド欠損マウスにおいて小脳の変性萎縮が著明に認められた。50週齢でプルキンエ細胞が約半数に減少し、100週齢になると約20%にまで減少することが判明した。電顕では、プルキンエ細胞の破壊像と細胞質における空砲、ミトコンドリアの変性が著明に認められた。これらの所見はマウスにみられた進行性の歩行障害とよく一致していた。小脳変性のメカニズム解明のため、小脳細胞の初代培養を確立し、特にプルキンエ細胞の数と突起伸長に注目して解析を進めた。 KOマウスのプルキンエ細胞は野生型の1/3くらいしか見られず、生後間もない時点での生存能力に著しい産量が見られた。さらに、複合型ガングリオリドを欠損するGM2/GD2合成酵素のKOマウスとb-シリーズガングリオリドを欠損するGD3合成酵素のKOマウスを交配して、ダブルノックアウトマウス(D-KO)を作成した。 GM3のみと発現するD-KOマウスは、幼若時より末梢神経の変性と破壊が認められ、12週齢頃より、顔面や頚部に難治性の皮膚損傷が出現した。はじめに比較的小規模の外傷が発生し、それを頻回に掻爬することで損傷部が拡大するものと推定された。その要因として、神経変性に基づく痛覚の低下が考えられた。
