食道・胃の前癌性病変及び癌におけるSAGEを用いた網羅的トランスクリプトーム解析

研究概要

食道・胃の前癌性病変及び癌における遺伝子発現解析を通した癌の発生・進展の分子機構の解明を目的として、引き続いてSAGE法を用いて網羅的トランスクリプトーム解析を行った。前年度に同定したgenerating gene typeIV (RegIV)については、RT-PCR法による解析の結果、肺癌、乳癌、肝癌、食道癌では殆ど発現は認められず、定量的RT-PCR法においてステージの進んだ胃癌で高発現が認められ、さらに、in situ hybridization (ISH)法においても進行胃癌の浸潤部に特に強い発現が確認された。また、胃癌の原発巣とリンパ節転移巣において発現の異なる遺伝子をSAGE法により解析し、転移巣において発現が消失あるいは著しく低下していた3つのtag/遺伝子を抽出した。それらは、CDKインヒビターのp21、γCOPおよび未知のtagであった。γCOPは、粗面小胞体とゴルジ装置間の小胞輸送に関与するCOP I複合体の1サブユニットである。γCOPの発現は、正常のリンパ節や脾臓においても認められたため、laser capture microdissection後に原発巣と転移巣での発現をRT-PCR法で比較すると、明らかに転移巣において発現は減弱していた。これら遺伝子の前癌性病変における発現解析は、ISHによって進め、SAGE解析のために試料を収集中である。一方、び漫浸潤型のスキルス胃癌で特異的に高発現を示し、SAGE databaseにおいてマッチする遺伝子が同定されないtag(SES1,SES3,SES4)を見い出した。新規の胃癌特異的浸潤関連遺伝子である可能性があり、reverseSAGE法および5'RACE法によりクローニングを行っている。
