ガレクチン9の機能解析と結合分子の精製

研究概要	本研究の目的は我々が好酸球遊走因子として発見、クローニグしたエカレクチン、すなわちガレクチン9の生物反応における重要性を検討するため、1)ガレクチン9の生物学的作用の解析、2)ガレクチン9産生遊離機構の解明、3)ガレクチン9結合分子の発現と機能の検討することにある。平成13年度に明らかにされたことは、1)ガレクチン9は好酸球活性化因子として既知の因子群とは異なるユニークな活性化因子と考えられることをJ. Immunologyに報告した。さらに遊走活性の発現にはリンカーペプチドは関与していないことをGlycobiologyに報告した。好酸球以外の細胞におけるガレクチン9の生物学的作用の検討を行ったが、ガレクチン9は腫瘍細胞(乳癌及びメラノーマ)の凝集とアポトーシスを誘導する活性が存在すること、また腫瘍細胞における発現によって転移予知診断が出来ることをさらに確認できた(論文投稿中)。2)ジャーカットT細胞株におけるガレクチン9の産生・遊離はPMAによって惹起されるが、ガレクチン9の遊離にはMMPの関与が必須であることをGlycobiologyに報告した。さて、ガレクチン9は生体内においてはマスト細胞やマクロフナージが保有していることを明らかにした。現在、マスト細胞やマクロファージにおけるガレクチン9の産生・遊離に関して研究を展開中である。3)種々の細胞表面のガレクチン9結合分子はラクトースに対する親和性において異なる分子が存在することやそれらの分布は細胞によって異なることを明らかにした。さらにガレクチン9投与によって低親和性の結合分子を有する細胞がアポトーシスを誘導されることを明らかにした(論文投稿中)。現在、アポトーシスに関与する結合分子の精製と構造決定の研究を進展中である。
