低酸素適応における回虫ミトコンドリア特異的呼吸酵素群の生理機能

研究概要	われわれは回虫をモデル系として寄生虫における低酸素適応機構の研究を進め、ミトコンドリアのNADH-フマル酸還元系が宿主体内の嫌気的環境で中心的な役割を果たしている事を明らかにした。その生理的意義は嫌気的グルコース分解系の最終ステップとして、無酸素下でも複合体Iの共役部位を駆動する事によりATPを合成できる点にある。そこで本研究ではNADH-フマル酸還元系の分子構築とその生理機能の特徴を明らかにする目的で、回虫成虫ミトコンドリアを用い、本年度は以下の3点についての成果を得た。 (1)複合体I(NADH-ユビキノン還元酵素)について:回虫成虫ミトコンドリアを用い、複合体Iの詳細な酵素学的解析を行ない、キノン類や阻害剤に対する相互作用が宿主哺乳類と大きく異なる点を明らかにした。特にわれわれが見い出した線虫複合体Iの特異的阻害剤ナフレジンがロドキノンの結合部位を競合阻害する点を明らかにした。 (2)複合体II(ロドキノール-フマル酸還元酵素 ; RQQFR)について:成虫ミトコンドリアからRQQFRを界面活性剤を用いて可溶化し、イオン交換クロマトグラフィーで大量に精製する方法を確立し、結晶化条件の検討に入った。現在、微小ではあるが結晶が得られる条件を見い出した。 (3)ロドキノンおよびユビキノン生合成機構と生理的役割:自由生活性線虫Caenorhabditis elegansの長寿命変異株clk-1のキノン組成を分析した結果、clk-1ではユビキノンが全く合成されておらず、代わりに合成中間体であるデメトキシユビキノンが蓄積している事が明らかになった。しかし、ロドキノンの合成量に変化はないので、ユビキノンとロドキノンの合成経路の分岐はデメトキシユビキノンの上流である事が判明した。
