研究概要	1)樹状細胞上に発現するクラス4セマフォリンSema4Aを単離し、そのリコンビナント可溶性分子や抗sema4Aモノクローナル抗体を作成した。抗Sema4Aを用いたフローサイトメトリー解析から、Sema4Aは樹状細胞やB細胞に恒常的に発現するが、休止期のT細胞には発現していないことがわかった。リコンビナント可溶性Sema4Aは抗CD3抗体と抗CD28抗体によるT細胞の増殖や、エフェクター細胞への分化を有意に亢進した。また、抗Sema4A抗体はin vitroのMLR反応を抑制するとともに、in vivoに投与した場合も抗原特異的T細胞の出現を阻害するだけでなくMOG誘導性の実験的自己免疫性脳脊髄炎の発症をも抑えた。すなわちSema4Aは、T細胞の活性化分化に必須の分子であることが明らかになった。 2)種々の細胞に対するリコンビナント可溶性Sema4Aの結合を解析したところ、活性化T細胞とEL-4などのT細胞株の表面に、Sema4A受容体が発現していることが明らかとなった。次に、EL-4細胞から発現型cDNAライブラリーを作成し、発現クローニングを行い、Timファミリーに属するTim2をSema4A結合タンパクとして単離した。可溶性Sema4AはTim2発現細胞には結合したが、Timファミリーの他のメンバーを発現する細胞には結合しなかった。また、可溶性Tim2と可溶性Sema4Aの分子間結合も表面プラズモン共鳴法を用いて確認した。さらに、Tim2発現細胞を可溶性Sema4Aで刺激すると数分以内にTim2の細胞内領域のチロシンリン酸化が誘導された。これらの結果から、Tim2はSema4Aの活性化シグナルをT細胞に伝達していると考えられる。
