IL-18で誘導されるMyD88非依存在、IL-4依存性IgE産生誘導機構の解明

研究概要	IL-18はIL-12の共存下でT細胞、B細胞、NK細胞に作用して、IFN-γの産生を極めて強力に誘導する。一方、IL-2の存在下でIL-18はナイーブT細胞に作用して、IL-4とIL-13の産生ならびにCD40Lの発現を誘導する。IL-18のシグナルはMyD88/IRAK/TRAF6等のシグナル伝達分子を介して核内に伝達される。従って、MyD88KOマウスの脾細胞をin vitroでIL-18で刺激してもNK活性あるいはIFN-γの産生は誘導されなかった。また、IL-12とIL-18をMyD88KOマウスに投与しても、IFN-γの産生は誘導されなかった(WTマウスでは大量のIFN-γが検出されていた)。興味あることには、IL-18を投与したMyD88KOマウスでは、その血清中に早い時期から大量のIgEを検出することができた。しかし、前もってCD4^+T細胞を抗体処置で消去しておくと、IL-18を投与してもIgE産生は誘導されなかった。また、抗IL-4抗体と抗IL-4R抗体を投与したMyD88KOマウスにIL-18を投与してもIgE産生は起こらなかった。以上のことから、IL-18をMyD88KOマウスに投与すると、MyD88非依存的にCD4^+T細胞が刺激を受けてIL-4とCD40Lを発現すること、そしてCD40LとIL-4がB細胞に作用してIgE産生を誘導することが明らかとなった。しかし、IL-18をMyD88KOマウスに投与した場合、同様にIL-18を投与されたWTのマウスに比し、IL-4とIL-13の産生は軽度であった。また、今回の研究から、MyD88KOマウスでは、既にTh2優位に傾いており、SPFで飼育していてもIgEのレベルが高値であることが示された。今後は、IL-18のIL-4産生に結びつくシグナルの伝達経路を解明していきたいと考えている。
