記憶B細胞選択、維持を司るシグナルと分子設計の解明

研究概要	1.これまでにFas不全(lpr)マウスでは、高親和性B細胞クローンの選択の遅延が起こり、また記憶B細胞にはFasが胚中心B細胞の選択と記憶B細胞の構成に関与することを明らかにした。lprマウスで認められた異常が、B細胞のFasの機能不全を反映するか否かを明らかにするため、lpr及び正常マウス脾臓よりB, T細胞を精製し、lpr B細胞と正常B, T細胞をRAG-1欠損マウスに移入した。移入後抗原刺激を行い産生される記憶B細胞の数と蓄積する体細胞変異の数を解析した、この結果、同一環境下においてlpr記憶B細胞の細胞数及び蓄積する体細胞変異の数は正常B細胞と比較し著明に上昇し、これらの結果はB細胞上のFasを介したシグナルで記憶B細胞の産生が制御されている可能牲が明らかにされた。 2.胚中心反応において胚中心B細胞から記憶B細胞が分化すると考えられているが、その機構は不明である。我々は胚中心B細胞活性維持と記憶B細胞分化においてrasが関与するか否かを明らかにする目的で、優性抑制型H-rasを強発現するトランスジェニックマウスを免疫し、免疫後の胚中心及び記憶B細胞の産生とそれぞれに蓄積する体細胞産生の数とパターンを解析した。この結果、優性抑制型rasの発現により胚中心B細胞の産生が強く抑制されたが、記憶B細胞のレベルは正常マウスとほぼ同じレベルであった。一方、各B細胞に蓄積する体細胞変異の数は正常及びトランスジェニックマウス間で同レベルであった。これらの結果は、胚中心B細胞活性と記憶B細胞への分化が異なった機構により誘導されることを示唆する。
