B細胞活性と免疫記憶の形成に関わる分子機構の解明

研究概要	記憶B細胞維持におけるFasの役割を明らかにする目的で、正常およびFas不全C57BL/6マウスをNP-CGで免役し、免疫後40日目に記憶B細胞を精製し、あらかじめCG蛋白で免役した同系のマウスに移入した。移入後直ちに免疫すると高いレベルの高親和性IgG抗体による二次免疫反応が惹起される。しかし、正常マウス由来記憶B細胞を移入し10日放置後免疫する二次免疫反応のレベルは著明に低下し、さらに3週間放置すると二次免疫反応の誘導はまったく認められず、記憶B細胞はこの環境下で細胞死に陥った可能性が示唆された。しかし、Fas不全記憶B細胞を移入し10日あるいは3週間放置し免疫すると、二次免疫反応の誘導はいずれも同様に認められ、この結果は、正常記憶B細胞はFasを介したシグナルにより細胞死に陥った可能性が示唆された。正常マウス由来記憶B細胞を移入後に坑Fasリガンド(FasL)抗体を投与すると、移入後10日放置しても二次免疫反応の誘導は認められ、この結果はFas-FasLを介したシグナルにより記憶B細胞の維持が抑制される可能性が示唆された。これらの結果は、記憶B細胞の維持に、Fasを介したシグナルとそれに拮抗する未知のシグナルが関与する可能性を示唆する。記憶B細胞の生存に関与する因子を明らかにする目的で、記憶B細胞に強発現する遺伝子をクローニングした。この結果得られた遺伝子の内3つのクローンが細胞の生存維持に関与する可能性が示唆された。しかし、いずれの遺伝子をB細胞株へ強発現させても、Fasを介したアポトーシスへの感受性が低下することはなかった。Fasを介したシグナルとそれに拮抗する未知のシグナルを今後さらに異なったシステムを用いて探索する予定である。
