脂肪細胞における活性酸素の役割と肥満:細胞外SODを中心として

研究概要

1.白色脂肪組織(WAT)は、余剰エネルギーの貯蔵器官としてだけでなく、脂質代謝やエネルギー代謝に重要な役割をもち、レプチンやPAI-I(plasminogen activator inhibitor-1)、TNFα、細胞外分泌型SOD(EC-SOD)などの生理活性物質を分泌する内分泌器官として、生体におけるさまざまな役割が明らかになってきた。
マウス3T3-L1細胞は、インスリンなどの刺激により細胞内に脂肪滴を蓄積して脂肪細胞様に分化するモデル系としてよく用いられている。この細胞にアダプタータンパク質SH2-Bの野生型及び変異型cDNAを発現させたところ、脂肪細胞への分化に変化がみられた。すなわち、野生型のSH2-Bbを発現させた細胞では、通常のtime course(インスリン刺激後6・8日)でほとんどの細胞が脂肪滴を貯えて脂肪細胞に分化したが、一方、SH2ドメインに変異を導入したdominant negative型のSH2・Bbを発現させた細胞では脂肪細胞に分化したものはわずかであった。これらのことは、SH2・Bがインスリン受容体の下流で、脂肪細胞への分化のシグナル伝達に重要な役割を果たしている可能性を強く示唆する。
2.EC-SODがWAT、褐色脂肪組織のいずれにも核に局在することを免疫組織染色を用いて明らかにした。EC-SODのヘパリン結合ドメイン(C末端近傍)の一次構造(-R-K-K-R-R-R-)が核タンパク質の核移行シグナル(NLS)によく似ていることから、EC-SODの核移行へのこのドメインの関与を検討した。
マウスEC-SOD遺伝子のN末端(分泌シグナル)、あるいはC末端を改変したミュータント遺伝子を作成し、COS細胞を用いた一過性発現系で、ヘパリン結合ドメインが核移行シグナルとして働き、EC・SODは選択的に核へ運搬されていることを確認した。
