研究概要

1.SLE末梢血T細胞株より得た全mRNAを同胞非SLE T細胞全mRNAを比較し、前者に特徴的に発現するmRNAを追求した。その結果、2クロンを得た。これらのクロンにつき発現系を作成し、抗DNA抗体との結合性を検討した。しかし、特異的な結合は得られなかった。
2.SLE例末梢リンパ球のcDNAライブラリーを作成した。このcDNAライブラリーを発現させ、上記の抗DNA抗体O-81と反応しうるペプチドを検索した。プローブとしてO-81VH遺伝子を用いて大腸菌PinPoint^<TM>蛋白発現システムを用いたO-81scFVを作成した。O81scFVはssDNAと結合し、かつO-81抗イディオタイプ抗体とも結合し、O81抗体活性を有することが判明した。O-81scFVとSLEcDNAライブラリーより発現したクロンの結合をtwo-hybrid法にて調べたところ、16の反応クロンを得た。この中からO-81scFVに結合性の高いクロンを選択した。このクロンよりGST蛋白発現ベクターに組み込みリコンビナント蛋白を作成した。
3.同定された蛋白につき、生物活性、SLEmRNAでの発現における特異性等について検索した。
