動脈硬化形成過程における血管内皮細胞のアポトーシスの意義の解析

研究概要	アンジオテンシンII(ATIIは動脈硬化巣における血管平滑筋の脱分化、肥大に重要な役割を果たしている。しかしATIIどのような細胞内シグナル伝達経路/転写因子を介して血管平滑筋の脱分化、肥大を起こすのか不明の点が多い。一方心肥大ではcalcineurin/nuclear factors of activated T cells-3(NFAT3)、AP-1等の関与が示されているが、この経路の血管平滑筋肥大への関与に関しては不明である。そこで本研究ではATIIよりこれらの転写因子が活性化されるか否か、またこれらの転写因子が血管平滑筋の脱分化、肥大へ関与するかどうかを検討した。1)ATII刺激によりラット大動脈由来培養血管平滑筋細胞(VSMCs)のMEF2A蛋白の発現は刺激後8時間より約2.2倍に増加した。一方MEF2AのmRNAの発現量は変化しなかった。2)ATII刺激によりラットVSMCsでのMEF2AおよびNFAT3のDNA結合活性はそれぞれ2.0倍、2.4倍に増加した。3)ATII刺激によりヒトnonmuscle myosin heavy chain Bプロモーター活性は2.0倍に有意に増加し、この増加は10^<-6> M cyclosporin A投与により48%に抑制された。またATII刺激によりc-junプロモーター活性は3.0倍に有意に増加し、この増加はMEF2Aのdominant negative mutantのcotransfectionによりbaselineまで抑制された。以上よりATIIによりMEF2AおよびNFATを介する転写が亢進することが示された。ATIIよる血管平滑筋の脱分化、肥大は少なくとも一部はこれらの転写因子を介する可能性が示唆された。
