血管平滑筋細胞の可塑性を基盤とする動脈硬化発症の分子機構解明に関する研究

研究概要	血管平滑筋細胞の分化特性を解明するため血管平滑筋細胞特異的転写装置を解析し、分化型血管平滑筋細胞培養系を用いて脱分化因子の検索と血管内膜肥厚モデルラットを作製し、動脈硬化発症機転を追究した。カルデスモン・ミオシン重鎖・平滑筋型α-アクチン・SM22α・カルポニン・β-トロポミオシン・α1インテグリンなど平滑筋細胞特異的遺伝子の血管平滑筋細胞転写装置として、Nkx3.2(NKホメオ転写因子)・SRF・GATA6の存在を明らかにした。また、血管平滑筋細胞の分化・脱分化シグナルのスイッチング分子がチロシン脱リン酸化酵素SHP-2であることを見い出した。血清は強力な血管平滑筋細胞脱分化能を示す。そこで、ヒト血清中の血管平滑筋細胞脱分化因子の検索を同上培養系を用いて行い、不飽和リゾホスファチジン酸(LPA)が血清中の主要な脱分化因子であることを同定した。飽和LPAに脱分化能見い出されず、血管平滑筋細胞脱分化は不飽和LPA特異的である。またこれら不飽和LPAは、酸化LDL(moxLDL)および動脈硬化巣においても高濃度検出された。不飽和LPAによる血管平滑筋細胞脱分化は、上述のERKとp38MAPK系の協調的活性化により誘導され、不可逆的である。さらに、頸動脈へのLPA投与により不飽和LPA特異的に著明な血管内膜肥厚を惹起し、ERKとP38MAPK阻害剤の前処置により不飽和LPAによる血管内膜肥厚は完全に抑制した。以上の結果は、in vivoにおいても不飽和LPAによる血管内膜肥厚はERKとp38MAPK系の活性化による平滑筋細胞脱分化によることを明らかにし、新たな血管内膜肥厚モデルを提示した。
